    I opened tile two in the flower shop and it takes me to the 3rd floor. I can't find a way to the 2nd floor.

    You have opened the Cellar door (Tile 6). As -Paaryna11 stated, Tile 2 requires Lavendar and gives you access to the 2nd Floor:
